含硫氨基酸对糖尿病大鼠窦房结损伤的保护作用

Protective role of sulphur-containing amino acids on sinoatrial node dysfunction in type 1 diabetes rat model

摘要:

目的 探讨补充半胱氨酸或牛磺酸对糖尿病大鼠窦房结损伤的作用。  方法 SD大鼠随机分为正常对照组(C)组、糖尿病(DM)组、糖尿病+半胱氨酸(DM+Cys)组和糖尿病+牛磺酸(DM+Tau)组。检测SOD和MDA含量;测定窦房结功能SNRT、CSNRT、SACT和IHR;检测窦房结SOD2、HO-1、NOX2、NOX4、HCN2、HCN4表达。   结果 与C组比较,DM组血SOD降低,MDA升高;SNRT、CSNRT和SACT延长,IHR减慢;窦房结SOD2、HO-1、HCN2和HCN4表达降低,NOX2和NOX4表达升高(P<0.05)。与DM组比较,DM+Cys和DM+Tau组血SOD升高;IHR增加;窦房结SOD2、HO-1、HCN2和HCN4表达升高,NOX2和NOX4表达降低(P<0.05)。   结论 补充Cys或Tau能降低STZ大鼠窦房结氧化应激损伤,增加HCN4和HCN2表达,改善窦房结功能障碍。

Abstract:

Objective To explore the effects of cysteine or taurine on sinoatrial node dysfunction in diabetic rats and the underlying mechanism. Methods SD rats were randomly divided into four groups:control group(C) diabetes mellits group(DM), diabetes mellits+cysteine group(DM+Cys), diabetes mellits+taurine group(DM+Tau). The blood content of SOD and MDA were assessed. The sinoatrial node function was detected by SNRT, CSNRT, SACT and IHR. We also measured the expression of SOD2, HO-1, Nox2, Nox4, HCN2 and HCN4. Results  Compared with C group, the levels of SOD decreased, whereas MDA increased in DM group. The IHR was reduced with the prolonging of SNRT, CSNRT and SACT. The DM group exhibited down-regulated expression of SOD2, HO-1, HCN2 and HCN4 but up-regulated expression of NOX2 and NOX4 (P<0.05). Compared with DM group, DM+Cys and DM+Tau group showed increased SOD, prolonged IHR, up-regulated expression of SOD2, HO-1, HCN2 and HCN4, down-regulated expression of NOX2 and NOX4 (P<0.05).  Conclusion Cysteine or taurine supplementation can reduce the oxidative stress, increased the expression of HCN2 and HCN4, improve the sinoatrial node dysfunction in diabetic rats.
